Function

The cytotoxic action of BPI is limited to many species of Gram-negative bacteria; this specificity may be explained by a strong affinity of the very basic N-terminal half for the negatively charged lipopolysaccharides that are unique to the Gram-negative bacterial outer envelope. Has antibacterial activity against the Gram-negative bacterium P.aeruginosa, this activity is inhibited by LPS from P.aeruginosa.

Activity summary

Neutrophil Bactericidal Protein BP30 also known as NGAL or LCN2 is a small protein with a mass of approximately 25 kDa. It is expressed chiefly in neutrophils but is also found in several other tissues including the liver lung and kidney. Mechanically BP30 plays a role in binding and transporting iron by sequestering bacterial siderophores which deprives bacteria of the iron essential for their growth. This activity is important for its function in the immune response.

Biological function summary

BP30 exerts antimicrobial effects by interfering with bacterial iron acquisition limiting bacterial growth and proliferation. It is not part of a complex; rather it functions as a monomer although it can form complexes with siderophores. Beyond its antimicrobial role BP30 also participates in cellular processes like apoptosis and inflammation. Its presence in various tissues points to its function as more systemically involved in defense mechanisms.

Pathways

BP30 interacts with several important biological pathways including the innate immune response and iron metabolism. Within the innate immune pathway it works alongside other antimicrobial peptides and proteins to enhance the body's first line of defense against pathogens. Additionally in the iron metabolism pathway BP30 modulates iron availability interacting with proteins such as transferrin and ferritin to balance iron homeostasis.

Associated diseases and disorders

BP30 has a notable connection with kidney disease and some bacterial infections. In kidney disease BP30 serves as a biomarker for acute kidney injury indicating elevated levels when the kidneys are damaged. Its link to bacterial infections results from its primary function of iron sequestration depriving bacteria of necessary nutrients. BP30 is connected to hepcidin through its role in iron regulation which can influence the progression of these conditions.
