Cell adhesion, ankyrin-binding protein which may be involved in neurite extension, axonal guidance, synaptogenesis, myelination and neuron-glial cell interactions. Isoform 2/isoform 3 may be responsible for mediating and signaling axon-glial interaction during the early stages of myelination.

Nfasc186 also known as Neurofascin-186 is an important isoform of the Neurofascin protein with an approximate mass of 186 kDa. This protein is expressed mainly in the nervous system particularly at the nodes of Ranvier and axon initial segments where it plays a role in maintaining axonal integrity and facilitating rapid signal conduction. By anchoring ion channels to the neuronal membrane Nfasc186 is essential for the proper function and structure of neurons. This interaction allows the effective propagation of nerve impulses and directly influences signal transduction efficiency in the central and peripheral nervous systems.
Nfasc186 interacts with the cytoskeletal scaffold and forms part of a larger molecular complex that stabilizes nodes of Ranvier. It associates with proteins like ankyrin G and voltage-gated sodium channels contributing to electrical impulse propagation along the axon. By anchoring these components Nfasc186 preserves node structure and ensures rapid saltatory conduction. Its participation in maintaining the functional architecture of the neuronal environment highlights its importance in neural signaling and homeostasis.
Nfasc186 participates in the nodes of Ranvier assembly and maintenance pathway. This pathway is critical for nervous system communication and involves associated proteins such as Caspr and Contactin which work with Nfasc186 to stabilize the node structure and ensure axon insulation. By facilitating the clustering of ion channels at the nodes Nfasc186 supports efficient nerve impulse transmission and plays an integral role in neurotransmission processes across neural tissues.
Defects in Nfasc186 have links to neurological conditions like multiple sclerosis and hereditary spastic paraplegia. Both conditions involve disruptions in myelination and neuronal transmission. In multiple sclerosis the loss or dysfunction of Nfasc186 and related proteins such as ankyrin G may contribute to demyelination and nerve fiber damage. In hereditary spastic paraplegia altered function of the associated pathways involving Nfasc186 may lead to compromised axonal transport and spasticity. Understanding these connections highlights the importance of Nfasc186 in maintaining neurological health and its potential as a therapeutic target.
