ChIP-sequencing - Anti-NFIL3 antibody [EPR27211-70] (AB309508)
Chromatin was prepared from HepG2 cells. Cells were fixed with 1% formaldehyde for 10 minutes. ChIP was performed with 107 cells and 8 µg of ab309508 [EPR27211-70]. ChIP DNA was sequenced on the Illumina NovaSeq 6000 to a depth of 30 million reads. The Input control is also shown.

NFIL3 was immunoprecipitated from 0.35mg HepG2 (human hepatocellular carcinoma epithelial cell) whole cell lysate with ab309508 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab309508 at 1/1000 dilution (0.508 μg/ml). VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution. Lane 1 : HepG2 (human hepatocellular carcinoma epithelial cell) whole cell lysate (10 μg) Lane 2 : ab309508 IP in HepG2 (human hepatocellular carcinoma epithelial cell) whole cell lysate Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab309508 in HepG2 whole cell lysate Blocking and dilution buffer and concentration : 5% NFDM/TBST. Exposure time : 93 seconds. Observed band : 60 kDa Lysates were freshly made and used for Western blotting immediately to minimize protein degradation.
