Function

Component of a ribosome-associated translocon complex involved in multi-pass membrane protein transport into the endoplasmic reticulum (ER) membrane and biogenesis (PubMed:32820719). May antagonize Nodal signaling and subsequent organization of axial structures during mesodermal patterning, via its interaction with NOMO (By similarity).

Supplementary info

Biological function summary

Nicalin acts as a regulator of Notch signaling an essential pathway for cell differentiation and development. It is a part of Nicalin-Nomo1 complex playing a significant role in modulating protein interactions. This complex contributes substantially to modifying cell fate decisions especially in the nervous system. Nicalin's presence in this complex highlights its importance in fine-tuning signaling mechanisms.

Activity summary

Nicalin also known as NOMO1 is a protein involved in the Notch signaling pathway and has an approximate molecular mass of 70 kDa. This protein localizes predominantly in the endoplasmic reticulum. Nicalin expresses across multiple tissue types but shows higher levels in neural tissues. It interacts with Nomo1 and other proteins to form a multi-protein complex that modulates signaling functions.

Pathways

Nicalin's function strongly influences Notch and Wnt signaling pathways. These pathways are important for processes such as cell proliferation and apoptosis. In the Notch pathway Nicalin forms interactions with the Presenilin protein influencing the maturation of Notch receptor proteins. This interaction affects cellular communication highlighting Nicalin's integrative role within signaling networks.

Associated diseases and disorders

Nicalin shows a connection to neurodegenerative diseases like Alzheimer's Disease where its interaction with Presenilin is significant. The protein also relates to developmental disorders influencing nervous system development due to its role in the Notch signaling pathway. Nicalin's regulation within these pathways can impact disease progression and severity indicating its potential as a therapeutic target.

We have systematically measured KD (the equilibrium dissociation constant between the antibody and its antigen), of more than 840 recombinant antibodies to assess not only their individual KD values but also to see the average affinity of antibody.
Based on the comparison with published literature values for mouse monoclonal antibodies, Recombinant antibodies appear to be on average 1-2 order of magnitude higher affinity.
