Niemann Pick C1 Like 1 (NPC1L1) also known simply as NPC1L1 is a protein that plays a critical role in the intestinal absorption of dietary cholesterol. NPC1L1 is a multi-pass membrane protein located in the brush border of small intestine enterocytes and also present in the liver. It consists of approximately 1334 amino acids giving it a mass of around 148 kDa. This protein serves as a conduit for cholesterol as it moves across the cell membrane taking part in the cholesterol uptake mechanism.
Biological function summary

NPC1L1 is essential for maintaining cholesterol homeostasis within the body. It is not part of a larger protein complex but works closely with other proteins to facilitate lipid incorporation into chylomicrons. Inhibitors of NPC1L1 such as ezetimibe effectively reduce blood cholesterol levels by blocking this absorption route. The presence of NPC1L1 on the apical surface of enterocytes emphasizes its function in the body's cholesterol metabolism impacting systemic lipid levels.
