Ninein also known as NIN or Antiabrin is a centrosomal protein important for microtubule anchoring. It has a molecular mass of approximately 230 kDa. This protein is expressed in several tissues with heightened levels in nerve and epithelial cells. Ninein localizes primarily at the centrosome and is an important centrosome marker aiding in the attachment of microtubules at the centrosome during cell division.
Ninein is critical for proper cell cycle progression and cellular organization. It acts as a scaffolding protein playing a significant role in centrosome cohesion and microtubule anchoring by interacting with other centrosome proteins. Ninein is also a part of the pericentriolar matrix a component of the centrosome necessary for microtubule nucleation and stabilization ensuring the correct formation and orientation of the mitotic spindle.
Ninein plays a significant role in centrosome-related signaling pathways including microtubule organization and maintenance. It interacts with several proteins such as rootletin influencing centrosome cohesion and dynamics. Ninein is also implicated in the cell cycle regulation pathway where it works alongside pericentrin to maintain centrosome integrity and support chromosomal segregation.
Ninein has been linked to ciliopathies and some neurological disorders. In ciliopathies Ninein dysfunction can lead to improper cilia formation and function highlighting its role in cilia biogenesis. Moreover disruptions in Ninein expression or function can contribute to brain development disorders as it is involved in neuronal differentiation. Proteins such as centrin are often studied alongside Ninein to better understand its involvement in these conditions.
