Immunocytochemistry/ Immunofluorescence - Anti-Ninein/NIN antibody - Centrosome Marker (AB4447)

IF using ab4447 to detect Ninein/NIN in MCF7 cells (human).

The left hand panels are DAPI stained nuclei. The right hand panels are the same nuclei stained for Ninein/NIN.

ab4447 was used at 1/1000.

Ninein/NIN shows nuclear and centrosome staining, which is expected from the literature.

Immunocytochemistry/ Immunofluorescence - Anti-Ninein/NIN antibody - Centrosome Marker (AB4447)

ab4447 staining Ninein in HeLa cells. The cells were fixed with 4% PFA (10 mins), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab4447 (shown in green) at 0.1μg/ml and ab7291 (shown in red), Mouse monoclonal to alpha Tubulin, at 1μg/ml. The secondary antibodies used were ab150081, Alexa Fluor® 488 Goat anti-Rabbit IgG (H+L), and ab150120, Alexa Fluor® 594 Goat Anti-Mouse IgG (H&L) used at a 1/1000 dilution for 1h at room temperature. Nuclear DNA was labelled with DAPI (shown in blue).

Image was acquired with a high-content analyser (Operetta CLS, Perkin Elmer) and a maximum intensity projection of confocal sections is shown.

Product details

The centrosomal protein ninein has been identified as a microtubule minus end capping and centriole anchoring protein, but its true physiological function remains to be determined. At mitosis, ninein has been observed in association with the mitotic spindle as well as at the centrosomes.

Supplementary information

Ninein also known as NIN or Antiabrin is a centrosomal protein important for microtubule anchoring. It has a molecular mass of approximately 230 kDa. This protein is expressed in several tissues with heightened levels in nerve and epithelial cells. Ninein localizes primarily at the centrosome and is an important centrosome marker aiding in the attachment of microtubules at the centrosome during cell division.
Biological function summary

Ninein is critical for proper cell cycle progression and cellular organization. It acts as a scaffolding protein playing a significant role in centrosome cohesion and microtubule anchoring by interacting with other centrosome proteins. Ninein is also a part of the pericentriolar matrix a component of the centrosome necessary for microtubule nucleation and stabilization ensuring the correct formation and orientation of the mitotic spindle.

Pathways

Ninein plays a significant role in centrosome-related signaling pathways including microtubule organization and maintenance. It interacts with several proteins such as rootletin influencing centrosome cohesion and dynamics. Ninein is also implicated in the cell cycle regulation pathway where it works alongside pericentrin to maintain centrosome integrity and support chromosomal segregation.

Ninein has been linked to ciliopathies and some neurological disorders. In ciliopathies Ninein dysfunction can lead to improper cilia formation and function highlighting its role in cilia biogenesis. Moreover disruptions in Ninein expression or function can contribute to brain development disorders as it is involved in neuronal differentiation. Proteins such as centrin are often studied alongside Ninein to better understand its involvement in these conditions.

Target data

Centrosomal protein required in the positioning and anchorage of the microtubule minus-end in epithelial cells (PubMed : 15190203, PubMed : 23386061). May also act as a centrosome maturation factor (PubMed : 11956314). May play a role in microtubule nucleation, by recruiting the gamma-tubulin ring complex to the centrosome (PubMed : 15190203). Overexpression does not perturb nucleation or elongation of microtubules but suppresses release of microtubules (PubMed : 15190203). Required for centriole organization and microtubule anchoring at the mother centriole (PubMed : 23386061).
