NKIRAS1 also known as NFKB inhibitor interacting Ras-like 1 is a small protein with a mass of approximately 21 kDa. This protein belongs to the Ras superfamily of small GTPases and acts as a negative regulator. NKIRAS1 is primarily expressed in various tissues including the brain liver and heart. It prevents the degradation of IκB proteins therefore inhibiting NF-κB activation. Its main mechanical role is to bind and inhibit the phosphorylation and degradation of IκB a process involved in the activation of the NF-κB pathway.
Biological function summary

NKIRAS1 is involved in the regulation of immune and inflammatory responses. It forms a part of the IκB complex which prevents the nuclear translocation of NF-κB a transcription factor that controls the expression of several genes related to immune response and cell survival. As a result NKIRAS1 plays a role in maintaining cellular homeostasis and preventing excessive inflammation. By modulating NF-κB signaling it can influence processes like cell proliferation apoptosis and cytokine production.

Pathways

NKIRAS1 functions within the NF-κB signaling pathway and interacts with the TGF-β pathway. In the NF-κB pathway NKIRAS1 connects with proteins like IKK (IκB kinase) and IκB to control the release and activity of NF-κB. In the TGF-β pathway interactions with SMAD proteins have been noted suggesting a role in regulating cell growth and differentiation. These pathways highlight NKIRAS1's importance in immune regulation and cell fate decisions.

NKIRAS1 associates with inflammatory diseases and certain cancers. Abnormal expression or mutation of NKIRAS1 can lead to dysregulated NF-κB activity contributing to conditions like rheumatoid arthritis and various carcinomas. The protein's interactions with NF-κB and IκB make it a relevant target for therapeutic interventions in these diseases. Researchers are exploring modulation of NKIRAS1 to manage inflammation and inhibit cancer cell growth.

Atypical Ras-like protein that acts as a potent regulator of NF-kappa-B activity by preventing the degradation of NF-kappa-B inhibitor beta (NFKBIB) by most signals, explaining why NFKBIB is more resistant to degradation. May act by blocking phosphorylation of NFKBIB and mediating cytoplasmic retention of p65/RELA NF-kappa-B subunit. It is unclear whether it acts as a GTPase. Both GTP- and GDP-bound forms block phosphorylation of NFKBIB.
