Supplementary information

This supplementary information is collated from multiple sources and compiled automatically.

NLN also known as Neurolysin is a 77 kDa metalloendopeptidase enzyme with an ability to cleave neuropeptides at specific sites. Researchers frequently observe NLN in the central nervous system kidneys testis and muscle tissues. This enzyme notably exists as a mitochondrial matrix protein showing an evolutionary conservation in its structure across mammalian species. NLN plays a role in processing bioactive peptides displaying similarity in substrate specificity to its better-known counterpart thimet oligopeptidase.
Biological function summary

NLN degrades neuropeptides which aids in regulating their activity and maintaining homeostasis. It does not typically form part of a large complex but interacts directly with peptide substrates to exert its effects. By modulating peptide availability NLN influences neurological functions and potentially affects signaling pathways involved in mood pain and energy regulation. Researchers continue to explore NLN's physiological roles and interactions though its exact contributions outside peptide processing remain less detailed.

Pathways

Studies show NLN engages in the metabolism of bioactive peptides including enkephalins and tachykinins. In these pathways NLN acts to terminate peptide signaling by cleaving them which alters receptor interactions. Within these mechanisms NLN shares activity with proteins like neprilysin both contributing to peptide degradation but acting on different substrates or within distinct cellular locales. These pathways are critical for maintaining neural function and homeostasis underlying many processes in the nervous system.

Dysregulation of NLN activity links to conditions such as hypertension and Alzheimer's disease. In hypertension improper peptide degradation can affect blood pressure regulation. In Alzheimer’s disease NLN’s inability to regulate amyloid-beta peptides in association with neprilysin contributes to plaque accumulation a hallmark of the disease. Understanding NLN’s role offers potential therapeutic insights for correcting peptide imbalances related to these disorders.

Target data

Hydrolyzes oligopeptides such as neurotensin, bradykinin and dynorphin A (By similarity). Acts as a regulator of cannabinoid signaling pathway by mediating degradation of hemopressin, an antagonist peptide of the cannabinoid receptor CNR1 (By similarity).
