NLRP11 also known as NALP11 is part of the NOD-like receptor (NLR) family which includes proteins involved in immune system regulation. It weighs approximately 116 kDa. This protein is expressed in the cytoplasm of various tissues with higher levels detected in reproductive tissues such as the ovaries and in immune-related tissues. NLRP11 has a tripartite structure comprising a PYD domain a NACHT domain and leucine-rich repeats which enables it to participate in protein-protein interactions.
Biological function summary

NLRP11 engages in innate immune responses by modulating inflammatory signaling pathways. It is part of the inflammasome complex although its exact role within this complex is less understood compared to more studied members like NLRP3. NLRP11 may also influence cytokine release contributing to the body's defense against pathogens. The tissue-specific expression indicates its possible role in reproductive immunology and local inflammation.

Pathways

NLRP11 lies within the inflammasome pathway and is indirectly linked to the NF-kB signaling pathway. Within these it intersects with proteins such as ASC (Apoptosis-associated Speck-like protein containing a CARD) and caspase-1 highlighting its involvement in the regulation of inflammatory processes. The exact interactions within these pathways remain an active area of research but they suggest a potential regulatory role in cellular inflammation response.

NLRP11 has been tentatively connected to inflammatory and autoimmune conditions. Some studies suggest a possible involvement in conditions like systemic lupus erythematosus (SLE) due to its influence on cytokine regulation and inflammation. Moreover disruptions in its expression or function may relate to irregular immune responses in reproductive disorders. Further study is needed but NLRP11 shares connections with NLRP3 and other inflammasome components that are more firmly linked to such diseases.
