This data was developed usingab314498, the same antibody clone in a different buffer formulation. Blocking and diluting buffer and concentration : 5% NFDM/TBST Negative control : spleen(PMID : 21543645) Lane1 was freshly made and used for Western blotting immediately to minimize protein degradation. The bands beneath the target band (98 kDa) are expected to be degradation products (PMID : 33376129), with the exception of the band around 50kd. The identity of the bands around 50 kDa are unknown. In Western blot, Anti-GAPDH antibody [EPR16891] - Loading Control (ab181602) staining at 1/200000 dilution.

NLRP6 also known as NACHT LRR and PYD domain-containing protein 6 plays a role as a sensor in the immune system. This protein has a mass of approximately 109 kDa. It is expressed in multiple tissues with high levels in intestinal cells and immune-related tissues. NLRP6 functions as part of the inflammasome complex which is important for inducing inflammatory responses by activating caspase-1 and promoting the maturation of inflammatory cytokines such as IL-1β and IL-18.
Biological function summary

NLRP6 has significant roles in maintaining mucosal homeostasis and modulating the body's inflammatory responses. It forms part of the NLRP6 inflammasome complex which impacts immune homeostasis especially within the gut environment. Research indicates that NLRP6 influences both the regulation of the microbiota and the repair processes of the mucosal barrier making it essential for gut health and protection against infections.

Pathways

NLRP6 influences both inflammation and metabolic pathways offering insights into its broad function in immunity. It is a part of the inflammasome pathway which triggers the immune system to combat bacterial infections by activating cytokines. The protein interacts with other inflammasome components like ASC and caspase-1. Additionally NLRP6 is involved in the toll-like receptor (TLR) signaling pathway influencing signal transduction related to immune responses.

Defects in NLRP6 have been linked to inflammatory bowel diseases (IBD) and colitis. Studies show that alterations in NLRP6 function may disrupt gut microbiota balance leading to increased susceptibility to these diseases. Additionally altered NLRP6 activity can connect to colorectal cancer especially with proteins like NF-κB suggesting that its pathways play a role in tumorigenesis by affecting inflammation and immune responses.

Acts as the sensor component of the NLRP6 inflammasome, which mediates inflammasome activation in response to various pathogen-associated signals, leading to maturation and secretion of IL1B and IL18 (PubMed : 30392956, PubMed : 34678144). Inflammasomes are supramolecular complexes that assemble in the cytosol in response to pathogens and other damage-associated signals and play critical roles in innate immunity and inflammation (PubMed : 30674671). Acts as a recognition receptor (PRR) : recognizes and binds specific pathogens and other damage-associated signals, such as lipoteichoic acid (LTA), a cell-wall component of Gram-positive bacteria, or double stranded RNA (dsRNA) (PubMed : 30392956, PubMed : 33377178, PubMed : 34678144). May also recognize and bind lipopolysaccharide (LPS), a major component of the outer membrane of Gram-negative bacteria; however, LPS is probably not a major activator of the NLRP6 inflammasome (PubMed : 31932628, PubMed : 34678144). Following LTA- or dsRNA-binding, NLRP6 undergoes liquid-liquid phase separation (LLPS), enhancing multivalent interactions, an essential step for the formation of the NLRP6 inflammasome polymeric complex (PubMed : 34678144). The NLRP6 inflammasome acts by promoting recruitment of effector pro-inflammatory caspases (CASP1 and/or CASP4) that catalyze maturation and secretion of IL1B and IL18 in the extracellular milieu (PubMed : 12387869, PubMed : 30392956, PubMed : 30674671, PubMed : 34678144). The NLRP6 inflammasome plays a central role in the maintenance of epithelial integrity and host defense against microbial infections in the intestine (PubMed : 30392956). Required to restrict infection against Gram-positive bacteria by recognizing lipoteichoic acid (LTA), leading to recruitment of CASP4 and CASP1, and subsequent maturation and secretion of IL1B and IL18 (PubMed : 30392956, PubMed : 33377178). Involved in intestinal antiviral innate immunity together with DHX15 : recognizes and binds viral dsRNA to restrict infection by enteric viruses through the interferon pathway and GSDMD-dependent release of IL18 (PubMed : 34161762, PubMed : 34678144).
