Western blot - Anti-NMDAR1 antibody [EPR23397-66] - BSA and Azide free (Detector) (AB283847)
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab274377). Anti-NMDAR1 antibody [EPR23397-66] staining at 1/1000 dilution, shown in green; Mouse anti-GAPDH antibody [6C5] (ab8245) loading control staining at 1/20000 dilution, shown in red. In Western blot, ab274377 was shown to bind specifically to NMDAR1. A band was observed at 105-125 kDa in wild-type Neuro-2a cell lysates with no signal observed at this size in GRIN1 knockout cell line ab281960 (knockout cell lysate ab282987). To generate this image, wild-type and GRIN1 knockout Neuro-2a cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween®20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L 800CW and Goat anti-Mouse IgG H&L 680RD at 1/20000 dilution.

Product details
ab283847 is a BSA and Azide Free antibody supplied in an unconjugated format and it is suitable for sandwich ELISAs to quantify Mouse NMDAR1. The recommended pair for sandwich ELISA is:
Capture: ab283846, Mouse NMDAR1 Capture Antibody (unconjugated)
Detector: ab283847, Mouse NMDAR1 Detector Antibody (unconjugated)
The reference range value is 203.13 - 13000 pg/mL.
Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.
What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:
- - High batch-to-batch consistency and reproducibility
- - Improved sensitivity and specificity
- - Long-term security of supply
- - Animal-free batch production
For more information, read more on recombinant antibodies.
Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.
Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.
Sandwich ELISA
The recommended antibody orientation is based on internal optimization for ELISA-based assays. Antibody orientation is assay dependent and needs to be optimized for each assay type. Please note that the range provided for this antibody is only an estimation based on the performance of the product using the recommended antibody pair. Performance of the antibody pair will depend on the specific characteristics of your assay. We guarantee the product works in sandwich ELISA, but we do not guarantee the sensitivity or dynamic range of the antibody in your assay.

Biological function summary
NMDA receptors including the NMDAR1 subunit serve essential functions in synaptic plasticity and memory processes. As part of the receptor complex NMDAR1 contributes to the control of synaptic strength. It requires co-activation by glutamate and glycine or D-serine linking its function to excitatory neurotransmission. The receptor's capacity to modulate synaptic connections forms the basis of learning and long-term memory.
Pathways
NMDAR1 plays a critical role in the glutamatergic pathway influencing synaptic plasticity and memory function. It interacts with other proteins like PSD-95 within the postsynaptic density. NMDAR1 is also involved in the calcium signaling pathway due to its ability to facilitate calcium entry into neurons upon synaptic activity. This positions the receptor as an important modulator in pathways that control neuronal communication and plasticity.
