Blocking and diluting buffer and concentration : 5% NFDM/TBST.
Samples are non-boiled as boiling may cause protein aggregates.
Bands of 180KDa and 115KDa respectively represent the full length and endogenously cleaved form.
The expression profile observed is consistent with what has been described in the literature (PMID : 22523092)
Negative control : Liver (PMID : 10049997, 9712644).

Immunohistochemical analysis of paraffin-embedded mouse hippocampus tissue labeling NMDAR2B with ab254356 at 1/1000 (0.514 ug/ml) followed by a ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Positive staining on the mouse hippocampus (PMID : 7931566). The section was incubated with ab254356 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.

NMDAR2B was immunoprecipitated from 0.35 mg rat brain tissue lysate 10 μg with ab254356 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab254356 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/5000 dilution.

Biological function summary
The NMDAR2B subunit contributes to the regulation of synaptic strength and is essential for processes involved in learning and memory. As part of the NMDA receptor complex it mediates excitatory neurotransmission and is involved in synaptic plasticity processes such as long-term potentiation (LTP). These functions are significant for cognitive function and neural development. The receptor's role in signal transduction is aided by the unique properties conferred by the NMDAR2B subunit such as its high affinity for glycine and slower deactivation kinetics.
Pathways
NMDAR2B is involved in the glutamatergic signaling pathway which is important for neural communication. It also participates in the calcium signaling pathway affecting cellular responses to external stimuli. The protein interacts with CaMKII and PSD-95 which are proteins that influence synaptic strength and architecture through these pathways. Its involvement links it to a variety of signaling events important for brain function.
