N-myristoyltransferase 2 or NMT2 is an enzyme that catalyzes N-terminal myristoylation. It transfers myristic acid a 14-carbon saturated fatty acid to the N-terminal glycine residue of substrate proteins. This modification influences the protein's function stability and subcellular localization. NMT2 has a structure with an approximate mass of 53 kDa. It is expressed in various tissues including the liver and the brain contributing to diverse biological processes.
Biological function summary

NMT2 participates in the post-translational modification of proteins which impacts cellular signaling and protein-protein interactions. It exists within a myristoylation enzyme complex working alongside its isoform NMT1 which complements its activity. NMT2 influences pathways involving protein trafficking and cell growth playing a role in membrane localization and signal transduction of several key proteins.

NMT2 actively engages in the Wnt signaling and protein kinase C (PKC) pathways. These pathways contribute to regulating cell fate decisions and cellular responses to extracellular signals. In cancer abnormal NMT2 expression impacts cancer cell survival and metastasis. In neurological disorders changes in myristoylation affect synaptic function and neuronal signaling. NMT2 interacts with proteins like beta-catenin in cancer progression and synaptophysin in neurological conditions. Adds a myristoyl group to the N-terminal glycine residue of certain cellular and viral proteins (PubMed : 25255805, PubMed : 9506952). Also able to mediate N-terminal lysine myristoylation of proteins : catalyzes myristoylation of ARF6 on both 'Gly-2' and 'Lys-3' (PubMed : 32103017). Lysine myristoylation is required to maintain ARF6 on membranes during the GTPase cycle (PubMed : 32103017).
