ICC/IF image of ab70335 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab42905, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

IHC image of ab70335 staining in human normal kidney formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab70335, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

GST pull-downs with Pin1 mutants reveal two different types of binding proteins. (A) Large-scale GST pull-downs. One to two milligrams of nocodazole-treated HeLa lysates were incubated with GST fusion proteins bound to glutathione beads. Interacting proteins were run on a 5–12% gradient gel and stained with Coomassie Blue. (B) GST pull-downs were performed as in (A) with 1 mg of HeLa lysate. Interacting proteins were run on 10% SDS-PAGE, transferred to PVDF and blotted with the indicated antibody. To better compare the amount of fusion protein on the beads, 1/200 of each pull-down was run on a gel and stained with Coomassie Blue (PD). (C) Quantification of blots shown in (B). This protein has a mass of approximately 54 kDa and is part of the Drosophila behaviour/human splicing (DBHS) protein family. It is ubiquitously expressed across different tissues with particular abundance in the nucleus showing its importance in gene regulation.

P54nrb is a multifunctional protein critical in nuclear processes. It forms part of a complex involved in RNA processing and nuclear retention of edited RNA. This complex also includes the proteins PSF and PSPC1 contributing to its role in transcription regulation and RNA stability. p54nrb impacts gene expression through its ability to bind to DNA and RNA influencing numerous regulatory pathways within the cell.
