Nucleolar protein 1 (NOB1) plays an important role in ribosomal RNA processing and biogenesis. Often referred to as NOB1p it is a protein that weighs approximately 55 kDa. NOB1 is expressed mainly in the nucleolus. This protein contains several domains such as the PIN domain which is important in its RNA cleavage function.
Biological function summary

NOB1 functions as a part of a complex called the 26S proteasome regulatory complex. It participates in the maturation of the 20S proteasome facilitating the processing of 18S rRNA which is important for ribosome assembly. This role highlights its importance for maintaining ribosomal function and protein synthesis.

Pathways

NOB1's role in ribosome assembly ties it to the ribosome biogenesis and protein synthesis pathways. It works closely with proteins such as PNO1 and the 18S rRNA processing machinery. These interactions ensure that ribosomes are properly assembled and functional which is critical for cellular growth and division.

Mutations or dysregulation of NOB1 have implications in cancer development. Aberrant expression of NOB1 has been linked to tumorigenesis particularly in pathways involving the oncogenic protein MYC. By influencing ribosome biogenesis NOB1 can contribute to the uncontrolled proliferation characteristic of cancer cells.
