Supplementary information

NOC2L also known as Nucleus accumbens-associated protein 1 is a protein that plays a mechanical role in modulating nuclear functions. It has a molecular weight of approximately 107 kDa. NOC2L is expressed in various tissues including human central nervous tissues and several others related to developmental processes. This protein interacts with chromatin and affects gene expression suggesting its influence on cellular function and growth regulation.
Biological function summary

NOC2L controls activities related to transcription regulation. It does not appear to form a large multi-protein complex but works alongside other nuclear proteins. By repressing the transcription of specific gene sets NOC2L plays a role in cellular homeostasis and differentiation. It influences key developmental stages and cellular responses to external stimuli.

Pathways

NOC2L integrates into transcriptional regulatory networks. It is actively involved in the p53 signaling pathway which contributes to cellular responses to stress and DNA repair mechanisms. NOC2L interacts with proteins like MDM2 which are important in this signaling cascade. Additionally NOC2L influences pathways that pertain to chromatin modification reflecting its role in gene regulation.

NOC2L has links to cancer and developmental disorders. Aberrations in its expression or function have been associated with tumorigenesis likely due to its involvement in the p53 pathway. The modulation of transcriptional activity by NOC2L influences cancer cell proliferation and survival. It also relates to developmental disorders where alterations in gene expression during critical growth phases contribute to abnormal phenotypes. Interactions with the p53 protein further emphasize its role in maintaining cell integrity and responding to oncogenic events.

Target data

Acts as an inhibitor of histone acetyltransferase activity; prevents acetylation of all core histones by the EP300/p300 histone acetyltransferase at p53/TP53-regulated target promoters in a histone deacetylases (HDAC)-independent manner. Acts as a transcription corepressor of p53/TP53- and TP63-mediated transactivation of the p21/CDKN1A promoter. Involved in the regulation of p53/TP53-dependent apoptosis. Associates together with TP63 isoform TA*-gamma to the p21/CDKN1A promoter.
