NOL11 also known as Nucleolar Protein 11 plays a mechanical role in ribosome biogenesis. This protein is 77 kDa in mass and shows expression largely in the nucleolus of the cell. It participates in the proper processing of the 47S pre-ribosomal RNA (rRNA) into the mature 18S rRNA an essential step in the formation of functional ribosomes. NOL11 interacts with elements of the small-subunit (SSU) processome indicating its involvement in early stages of ribosome assembly.
Biological function summary

NOL11 contributes to cellular proliferation by ensuring efficient ribosome synthesis. It acts as a component of the SSU processome complex which is a large ribonucleoprotein complex essential for the maturation of pre-rRNA. The protein NOL11 serves as a critical factor required for the synthesis of rRNA a process that directly affects the production of ribosomes and subsequently protein synthesis influencing cell growth and division.

Pathways

The protein functions within two major biological pathways: ribosome biogenesis and the cell cycle. It has a central role in ribosome biogenesis impacting the processing of pre-rRNA and connecting with other rRNA processing factors like Utp21. NOL11 also influences the cell cycle by indirectly affecting cell growth through its impact on ribosome production tying in with growth control mechanisms that involve proteins like p53.

NOL11 shows associations with conditions such as North American Indian childhood cirrhosis and Diamond-Blackfan anemia. These connections arise due to its pivotal function in ribosome biogenesis; any impairment can disrupt cellular protein synthesis leading to such disorders. NOL11 interacts with proteins involved in these conditions like those in the Fanconi anemia related pathways which further establishes its role in disease pathogenesis.
