NOL5A also known as Nucleolar Protein 5A or hNop56 functions as a nucleolar protein involved in ribosome biogenesis. It is characterized by a mass of approximately 63 kDa. NOL5A exhibits high expression within the nucleolus of cells where it participates in the assembly of ribosomal subunits. Specifically NOL5A plays a role in the modification and maturation of pre-ribosomal RNA. Its expression is observed in rapidly dividing cells highlighting its importance in cellular growth and proliferation.
Biological function summary

Ribosome biogenesis depends on several proteins and NOL5A serves as a core component in the small nucleolar ribonucleoprotein (snoRNP) complex. This complex is responsible for the processing and chemical modification of small nucleolar RNAs (snoRNAs) which are essential for the production of functional ribosomes. NOL5A helps facilitate the cleavage and assembly steps necessary for pre-rRNA to mature into functional rRNAn important for effective protein synthesis within the cell. Its activity is important to maintaining proper ribosome function ensuring efficient protein translation.

Pathways

NOL5A acts within the rRNA processing pathway and is also linked to the mTOR signaling pathway. In the rRNA processing pathway NOL5A's role in the nucleoplasm involves cooperation with other nucleolar proteins like fibrillarin and Nop58 to orchestrate rRNA processing. In the mTOR pathway the regulation of ribosome production connects NOL5A to protein synthesis pathways impacting cell growth and proliferation. mTOR signaling influences many cellular processes and NOL5A’s function in ribosome biogenesis links it to these critical growth and survival pathways.

NOL5A has connections with cancer and ribosomopathy. Altered expression of NOL5A has implications in various cancers as its role in ribosome production can drive aberrant cell proliferation. Additionally proper function of NOL5A is essential in preventing ribosomopathies disorders characterized by defects in ribosome biogenesis. These disorders often result in bone marrow failure or craniofacial abnormalities. Studies investigating NOL5A's interactions with proteins such as NPM1 show its involvement in nucleolar stress responses which can influence cancer progression and response to therapy.

Involved in the early to middle stages of 60S ribosomal subunit biogenesis. Required for the biogenesis of box C/D snoRNAs such U3, U8 and U14 snoRNAs (PubMed : 12777385, PubMed : 15574333). Part of the small subunit (SSU) processome, first precursor of the small eukaryotic ribosomal subunit. During the assembly of the SSU processome in the nucleolus, many ribosome biogenesis factors, an RNA chaperone and ribosomal proteins associate with the nascent pre-rRNA and work in concert to generate RNA folding, modifications, rearrangements and cleavage as well as targeted degradation of pre-ribosomal RNA by the RNA exosome (PubMed : 34516797).
