Part of the small subunit (SSU) processome, first precursor of the small eukaryotic ribosomal subunit. During the assembly of the SSU processome in the nucleolus, many ribosome biogenesis factors, an RNA chaperone and ribosomal proteins associate with the nascent pre-rRNA and work in concert to generate RNA folding, modifications, rearrangements and cleavage as well as targeted degradation of pre-ribosomal RNA by the RNA exosome.

NOL6 also known as nucleolar protein 6 is a nucleolar RNA-associated protein with a mass of roughly 117 kDa. It localizes mainly to the nucleolus and shows high expression in actively dividing cells reflecting its role in ribosomal RNA (rRNA) processing. The NOL6 protein interacts with rRNA during ribosome biogenesis and its presence is notable in developing and differentiating tissues pointing to its involvement in fundamental cell growth and proliferation mechanisms.
NOL6 participates in ribosomal assembly by interacting with pre-ribosomal particles. It becomes part of the larger ribonucleoprotein complex important for the maturation of the 60S ribosomal subunit. NOL6 ensures the accurate cleavage and modification of precursor rRNA which is essential for functional ribosome construction. This contribution influences protein synthesis rates within the cell impacting overall cellular metabolism and responsiveness to growth signals.
Ribosome biogenesis and RNA processing in the nucleolus engage NOL6 extensively. It closely interacts with the rRNA processing pathway and is functionally connected to other nucleolar proteins such as fibrillarin and NOP56. HeLa (human epithelial cell line from cervix adenocarcinoma) cells stained for NOL6 (green) using ab228836 (1/500 dilution) in ICC/IF. Cells were fixed in 4% paraformaldehyde at RT for 15 minutes.
