NOL9 also known as Nucleolar Protein 9 is a protein encoded by the NOL9 gene. It has a molecular mass of approximately 150 kDa. This protein mainly localizes in the nucleolus a sub-region of the nucleus where it is highly expressed. NOL9 functions as a polynucleotide 5'-kinase which means it is involved in adding phosphate groups to the 5' end of RNA molecules. This activity plays an important role in processes such as RNA maturation.
Biological function summary

NOL9 participates in ribosome biogenesis which is the process of making ribosomes. It plays an important part in the processing and modification of precursor ribosomal RNA (pre-rRNA) into mature ribosomal RNA (rRNA) a task that is key for proper ribosome functionality. NOL9 functions within a multiprotein complex engaging with other nucleolar proteins to achieve efficient ribosomal RNA processing. The proper assembly and function of this complex ensure correct ribosome production a vital step for protein synthesis within the cell.

Pathways

NOL9 is involved in the rRNA processing pathway and ribosome assembly. It collaborates with other important proteins like fibrillarin and nucleolin to guide the transformation of pre-rRNA into functional rRNA components. This transformation is part of the ribosome biogenesis pathway which supports cellular growth and protein synthesis. The efficient functioning of NOL9 in these pathways helps maintain cellular homeostasis and protein production.

NOL9 has been related to potential implications in cancer and genetic ribosomopathy. Polynucleotide kinase that can phosphorylate the 5'-hydroxyl groups of single-stranded and double-stranded RNA and DNA substrates (PubMed : 21063389). Involved in rRNA processing and its kinase activity is required for the processing of the 32S precursor into 5.8S and 28S rRNAs, more specifically for the generation of the major 5.8S(S) form (PubMed : 21063389). Required for the efficient pre-rRNA processing of internal transcribed spacer 2 (ITS2) (PubMed : 21063389). Associates with LAS1L to form an ITS2 pre-rRNA endonuclease-kinase complex and is responsible for the transport of this complex into the nucleolus (PubMed : 31288032).
