Functions as a receptor for membrane-bound ligands Jagged-1 (JAG1), Jagged-2 (JAG2) and Delta-1 (DLL1) to regulate cell-fate determination. Upon ligand activation through the released notch intracellular domain (NICD) it forms a transcriptional activator complex with RBPJ/RBPSUH and activates genes of the enhancer of split locus (PubMed:21378985, PubMed:21378989). Affects the implementation of differentiation, proliferation and apoptotic programs (By similarity). Involved in bone remodeling and homeostasis. In collaboration with RELA/p65 enhances NFATc1 promoter activity and positively regulates RANKL-induced osteoclast differentiation (PubMed:29149593). Positively regulates self-renewal of liver cancer cells (PubMed:25985737).

Activity summary

Notch2 also known as neurogenic locus notch homolog protein 2 is a transmembrane receptor protein that plays a critical role in cell differentiation and development. It is part of the Notch signaling pathway which is highly conserved across species. The protein weighs approximately 270 kilodaltons and is primarily found in the kidney liver and various other tissues. Notch2 activation occurs through interactions with specific ligands leading to the release of its intracellular domain which enters the nucleus to regulate gene expression.

Biological function summary

Notch2 modulates important developmental processes including lineage specification and cell fate determination. This protein is vital in maintaining the homeostasis of several organs and tissues and it does this by being part of the Notch signaling complex. Additionally Notch2 contributes to hematopoiesis influencing the balance between different blood cell types. Researchers have also noted its role in maintaining stem cell populations and facilitating the proper function of the immune system.

Pathways

Notch2 integrates into the canonical Notch signaling pathway which impacts cellular communication and gene transcription. This pathway interlinks with the Wnt signaling pathway influencing tissue regeneration and cancer development. Associated diseases and disorders
