Notch2 also known as neurogenic locus notch homolog protein 2 is a transmembrane receptor protein that plays a critical role in cell differentiation and development. It is part of the Notch signaling pathway which is highly conserved across species. The protein weighs approximately 270 kilodaltons and is primarily found in the kidney liver and various other tissues. Notch2 activation occurs through interactions with specific ligands leading to the release of its intracellular domain which enters the nucleus to regulate gene expression.
Biological function summary

Notch2 modulates important developmental processes including lineage specification and cell fate determination. This protein is vital in maintaining the homeostasis of several organs and tissues and it does this by being part of the Notch signaling complex. Additionally Notch2 contributes to hematopoiesis influencing the balance between different blood cell types. Researchers have also noted its role in maintaining stem cell populations and facilitating the proper function of the immune system.

Pathways

Notch2 integrates into the canonical Notch signaling pathway which impacts cellular communication and gene transcription. This pathway interlinks with the Wnt signaling pathway influencing tissue regeneration and cancer development. Notch2 itself associates with proteins like Jagged1 and Delta-like ligands which are imperative to the signaling cascade. Cross-talk between these pathways highlights its importance in ensuring precise regulation of cellular functions and maintaining tissue integrity.

Notch2 has links to Alagille syndrome and certain malignancies like breast cancer. Mutations or dysregulation in the Notch2 signaling pathway can lead to these conditions. In Alagille syndrome for instance the interplay between Notch2 and Jagged1 mutations alters bile duct development leading to liver and heart problems. Also its role in various cancers suggests that altered Notch2 signaling contributes to uncontrolled cell proliferation and tumor progression.

Functions as a receptor for membrane-bound ligands Jagged-1 (JAG1), Jagged-2 (JAG2) and Delta-1 (DLL1) to regulate cell-fate determination. Upon ligand activation through the released notch intracellular domain (NICD) it forms a transcriptional activator complex with RBPJ/RBPSUH and activates genes of the enhancer of split locus (PubMed : 21378985, PubMed : 21378989). Affects the implementation of differentiation, proliferation and apoptotic programs (By similarity). Involved in bone remodeling and homeostasis. In collaboration with RELA/p65 enhances NFATc1 promoter activity and positively regulates RANKL-induced osteoclast differentiation (PubMed : 29149593).
