Supplementary information

Noto also known as Not or Notochord is a small nuclear protein with a mass of approximately 43 kDa. It serves a critical role in the notochord development acting as a transcription factor. Scientists often find it expressed in the primitive node and notochordal plate during embryogenesis. Its expression is necessary for the formation of body axis structures influencing the patterning of mesodermal tissues.
Biological function summary

The protein is involved in early vertebrate development impacting the proper development of the notochord which is essential for axial skeleton formation. Noto functions as part of transcriptional regulatory complexes that drive the expression of specific target genes. It interacts with other developmental regulators to ensure the differentiation and organization of midline structures.

Pathways

Noto plays a role in the Wnt and Hedgehog signaling pathways which are important for embryonic development particularly in establishing the body plan. It interacts with proteins like SHH (Sonic Hedgehog) and BMP (Bone Morphogenetic Protein) during these processes. These interactions contribute to the precise orchestration of signals required for tissue specification and the establishment of bilateral symmetry.

Dysregulation of Noto expression associates with congenital spine defects such as scoliosis and spina bifida. Abnormalities in Noto function also link with developmental disorders like caudal dysgenesis. Disruptions in its interaction with SHH can result in improper neural tube closure highlighting its significance in human developmental diseases.

Target data

Transcription regulator acting downstream of both FOXA2 and Brachyury (T) during notochord development. Required for node morphogenesis. Is essential for cilia formation in the posterior notochord (PNC) and for left-right patterning; acts upstream of FOXJ1 and RFX3 in this process and is required for the expression of various components important for axonemal assembly and function. Plays a role in regulating axial versus paraxial cell fate. Activates the transcription of ciliary proteins C11orf97 homolog, FAM183B and SPACA9 in the embryonic ventral node (By similarity).
