NOTUM is a lipase-like enzyme that functions mechanically to remove lipid modifications specifically palmitoleate from the Wnt proteins. It is alternatively known as palmitoleoyl-protein carboxylesterase. The molecular mass of NOTUM is approximately 53 kDa. NOTUM is expressed mainly in tissues such as the liver kidneys and gut where it plays a role in regulating signaling pathways.
Biological function summary

NOTUM inhibits the Wnt signaling pathway by cleaving the lipid moiety attached to Wnt proteins making it inactive. NOTUM acts independently and is not part of larger protein complexes. This regulation of Wnt signaling is important for maintaining cellular processes such as cell proliferation differentiation and migration especially during development and tissue homeostasis.
