NPHP1 also known as Nephrocystin-1 plays an important role in maintaining the function and structure of primary cilia. This protein has a molecular mass of approximately 79 kDa. NPHP1 is primarily expressed in the renal epithelial cells but it also shows expression in other tissues such as the eye and brain. The protein localizes to the cell's centrosome which acts as an important hub for microtubule organization and organelle trafficking.
Biological function summary

Nephrocystin-1 forms part of a ciliary protein complex involved in the signal transduction pathways that regulate cell proliferation and differentiation. Together with other proteins like INVS and NPHP4 Nephrocystin-1 contributes to the maintenance of ciliary function essential for sensing fluid flow in kidney ducts and other mechanosensory processes. The protein complex influences the positioning and anchoring of ciliary basal bodies contributing to ciliogenesis.

Pathways

Nephrocystin-1 participates in essential cellular signaling pathways particularly the Wnt signaling pathway and Hedgehog signaling. Through these pathways Nephrocystin-1 interacts with proteins such as Dishevelled and Gli impacting cellular processes like development migration and polarity. Disruptions in these pathways can lead to defects in tissue homeostasis underlining NPHP1's role in proper cellular function and response to external signals.

Mutations or malfunctions in Nephrocystin-1 can lead to nephronophthisis a form of hereditary kidney disease and Senior-Loken syndrome a disorder combining nephronophthisis with retinal degeneration. Nephrocystin-1 is closely linked with other ciliary proteins such as NEK8 and CEP290 which also participate in the pathological developments of these disorders. Effective functioning of NPHP1 and these associated proteins is essential for preventing the ciliary abnormalities observed in such ciliopathies.

Involved in the organization of apical junctions in kidney cells together with NPHP4 and RPGRIP1L/NPHP8 (By similarity). Does not seem to be strictly required for ciliogenesis (By similarity). Seems to help to recruit PTK2B/PYK2 to cell matrix adhesions, thereby initiating phosphorylation of PTK2B/PYK2 and PTK2B/PYK2-dependent signaling (By similarity). May play a role in the regulation of intraflagellar transport (IFT) during cilia assembly. Required for normal retina development (By similarity). In connecting photoreceptor cilia influences the movement of some IFT proteins such as IFT88 and WDR19. Involved in spermatogenesis (By similarity).
