Supplementary information

Nucleophosmin/Nucleoplasmin 2 (NPM2) also referred to as Nucleoplasmin-2 is a protein with molecular mass approximately 22 kDa. It belongs to the family of nucleoplasmins which are chromatin remodeling proteins. NPM2 is mainly expressed in oocytes and plays a significant role during early embryonic development. The protein is highly localized in the nuclei of developing oocytes and is known for its capacity to bind and exchange histones which is essential for chromatin structure.
Biological function summary

The function of NPM2 involves regulating chromatin in germ cells and early embryogenesis. It acts as a histone chaperone forming complexes with histones to facilitate chromatin assembly and disassembly. The protein enables the proper packing and unpacking of DNA ensuring genomic stability during cell division. NPM2 does not work alone and it often forms a complex with histone proteins allowing it to modulate chromatin dynamics efficiently.

Pathways

NPM2 participates in key cellular processes related to chromatin remodeling and regulation. It plays an important role in the chromatin assembly pathway working alongside proteins like histone H1 to modulate DNA accessibility. Additionally NPM2 interacts with other histone chaperones affecting the nucleosome assembly pathway. Through these pathways it influences various nuclear processes important for gene expression and maintenance of nuclear architecture.

NPM2 is associated with issues in reproductive health and cancer. Abnormal expression or malfunction of NPM2 can lead to infertility due to defects in oocyte maturation. In cancer biology variations in NPM2 expression levels might correlate with tumor progression where it interacts with several oncogenic proteins. For example changes in the function of NPM2 might influence the activity of histone-modifying enzymes linked to chromatin changes seen in malignant cells.
