Choline-specific phosphodiesterase that hydrolyzes sphingomyelin releasing the ceramide and phosphocholine and therefore is involved in sphingomyelin digestion, ceramide formation, and fatty acid (FA) absorption in the gastrointestinal tract (PubMed:12671034, PubMed:12885774, PubMed:15205117, PubMed:16255717, PubMed:28292932). Has also phospholipase C activity and can also cleave phosphocholine from palmitoyl lyso-phosphatidylcholine and platelet-activating factor (PAF) leading to its inactivation (PubMed:12885774, PubMed:16255717). Does not have nucleotide pyrophosphatase activity (PubMed:12885774). May promote cholesterol absorption by affecting the levels of sphingomyelin derived from either diet or endogenous sources, in the intestinal lumen (By similarity).

NPP-7 also known as nucleotide pyrophosphatase/phosphodiesterase 7 is an enzyme involved in the hydrolysis of pyrophosphate-containing substrates. It has a molecular mass of approximately 125 kDa. This enzyme exhibits expression in several human tissues prominently in the intestinal tract and kidney. The enzyme's activity facilitates the breakdown of molecules serving roles in both digestion and cellular signaling processes.
NPP-7 participates in lipid metabolism and contributes to maintaining homeostasis by degrading extracellular nucleotides. It forms part of larger protein complexes that regulate various cellular activities. Its enzymatic function is important for controlling the concentration of lipid-derived signaling molecules thereby impacting cellular communication and response mechanisms.
NPP-7 integrates into the pathway regulating phospholipid metabolism and nucleotide salvage pathways. It interacts with proteins such as NPP-2 which are involved in similar metabolic processes ensuring balance in lipid signaling and energy storage within cells. Its function in these pathways highlights its role in modulating phospholipid levels and influencing liver function and cellular energy dynamics.
NPP-7 relates to conditions such as metabolic syndrome and chronic kidney disease. Dysregulation or altered expression of NPP-7 can affect lipid metabolism and kidney function contributing to these diseases.
