The NPY2R also known as the Y2 receptor is a G-protein coupled receptor with a molecular mass of approximately 42 kDa. It primarily binds neuropeptide Y (NPY) peptide YY (PYY) and pancreatic polypeptide (PP). The NPY2R is expressed in the central nervous system particularly in the brain regions like the hypothalamus and hippocampus as well as in peripheral tissues such as vascular smooth muscle and the gastrointestinal tract.
Biological function summary

The NPY2R is involved in processes like appetite regulation anxiety and blood pressure control. When activated by its ligands NPY2R plays a role in reducing neurotransmitter release which translates into modulation of neuronal activity and vascular tone. It does not form part of a larger complex but functions by itself to exert its biological roles.

Pathways

NPY2R participates in the neuropeptide signaling pathway and is linked to the regulation of energy homeostasis. Through its interactions NPY2R influences the action of other receptors such as the Y1 receptor both contributing to the complex regulation of food intake and stress responses. The Y2 receptor also affects the sympathetic nervous system through the modulation of noradrenaline release.

NPY2R shows a connection with obesity and hypertension. Abnormal Y2 receptor function could lead to disrupted energy balance and increased appetite contributing to obesity. In hypertension altered NPY2R activity may lead to dysfunctional blood pressure regulation. Connections with proteins like Y1 receptor also underpin its relevance in these conditions as they together orchestrate the cardiovascular and metabolic responses.
