Neuregulin 1 (NRG1) also known as heregulin beta 1 is a member of the neuregulin protein family. It is a signaling protein with a molecular mass that typically ranges around 44 kDa. Researchers have found NRG1 expressed in various tissues including the heart brain and certain epithelial cells. It plays important roles in cell-cell communication by binding to ErbB family receptors particularly ErbB3 and ErbB4 which trigger downstream signaling processes.
Biological function summary

NRG1 is involved in several cellular activities such as cell growth survival and differentiation. It is not part of a complex but acts directly through its interactions with receptor tyrosine kinases in the ErbB receptor family. NRG1 influences cardiac development and neural function by mediating these cellular processes promoting the development and maintenance of neural and cardiac tissues.

Pathways

NRG1 is involved in the PI3K-Akt and MAPK/ERK signaling pathways. These pathways are critical for cellular growth differentiation and survival supporting the functions of NRG1 in various tissues. The interaction with ErbB2 and ErbB3 receptors is especially significant for these pathways as they help regulate the cellular responses initiated by NRG1 signaling.

NRG1 plays an impactful role in the development of certain types of cancer and schizophrenia. Dysregulation or mutations in the NRG1 gene can lead to altered signaling pathways which are connected to the pathogenesis of these conditions. In cancer the interaction with ErbB2 has been under investigation as it may promote tumorigenesis. In the context of schizophrenia NRG1's effects on neural development and neurotransmission are linked with the disorder's symptoms.

Direct ligand for ERBB3 and ERBB4 tyrosine kinase receptors. Concomitantly recruits ERBB1 and ERBB2 coreceptors, resulting in ligand-stimulated tyrosine phosphorylation and activation of the ERBB receptors. The multiple isoforms perform diverse functions such as inducing growth and differentiation of epithelial, glial, neuronal, and skeletal muscle cells; inducing expression of acetylcholine receptor in synaptic vesicles during the formation of the neuromuscular junction; stimulating lobuloalveolar budding and milk production in the mammary gland and inducing differentiation of mammary tumor cells; stimulating Schwann cell proliferation; implication in the development of the myocardium such as trabeculation of the developing heart. Isoform 10 may play a role in motor and sensory neuron development. Binds to ERBB4 (PubMed : 10867024, PubMed : 7902537). Binds to ERBB3 (PubMed : 20682778). Acts as a ligand for integrins and binds (via EGF domain) to integrins ITGAV : ITGB3 or ITGA6 : ITGB4. Its binding to integrins and subsequent ternary complex formation with integrins and ERRB3 are essential for NRG1-ERBB signaling. Induces the phosphorylation and activation of MAPK3/ERK1, MAPK1/ERK2 and AKT1 (PubMed : 20682778). Ligand-dependent ERBB4 endocytosis is essential for the NRG1-mediated activation of these kinases in neurons (By similarity).
