Activity summary

The Nik-related kinase (NRK) also known as MINK1 or Murine-inhibitor-of-apoptosis-protein-interacting-NK-1 is a protein kinase belonging to the germinal center kinase family. NRK has an approximate molecular mass of 195 kDa. This kinase expresses mostly in muscle tissues and the nervous system indicating its role in specific physiological processes related to these organs. NRK acts by phosphorylating target proteins which involves adding a phosphate group to specific serine or threonine residues influencing various signaling pathways.

Biological function summary

Activities of NRK affect cell signaling and dynamic structural rearrangement. As part of larger protein complexes NRK interacts with other kinases to regulate cytoskeletal organization and cell morphology. NRK also has a role in embryonic development contributing to muscle formation and neuronal differentiation. This positioning in cellular processes marks NRK as an essential component in developmental biology and tissue-specific anatomical pathways.

Pathways

NRK plays critical roles in signaling processes like the JNK/MAPK pathway. In this context NRK helps to mediate stress responses and apoptotic processes by interfacing with other kinases such as MKK4 and JNK1. NRK's activity influences cellular responses to environmental or cellular stress coordinating gene expression changes for appropriate cellular adaptation or apoptosis when necessary.

Associated diseases and disorders

Alterations in NRK are linked to the development of certain cancers including colorectal cancer and neurodegenerative disorders like Alzheimer's disease. Changes in NRK expression or function can disrupt normal cellular signaling contributing to tumor progression or neural tissue degeneration. For instance deregulation of the NRK-JNK pathway may involve a crosstalk with proteins like Tau which plays a significant role in Alzheimer pathology highlighting NRK's importance in maintaining cellular homeostasis.
