NSMase also known as neutral sphingomyelinase is an enzyme with a molecular mass typically ranging between 43 to 48 kDa. It plays an important role in the hydrolysis of sphingomyelin to generate ceramide and phosphocholine. nSMase is distributed widely in various tissues with higher expression levels observed in the brain liver and spleen. This enzyme exists in multiple isoforms each localized in different cellular compartments contributing to diverse cellular functions.
Biological function summary

Neutral sphingomyelinase influences several cellular processes by modulating ceramide levels. Ceramide a bioactive lipid serves as a secondary messenger in signal transduction pathways and mediates processes like apoptosis and cell differentiation. nSMase often functions as a monomer but it can also be part of larger complexes with other proteins to execute its roles in cellular signaling. Through its enzymatic activity nSMase impacts membrane structure and dynamics affecting overall cellular homeostasis.

Pathways

Neutral sphingomyelinase is involved in key signaling pathways like the MAPK and apoptosis pathways. In the MAPK pathway nSMase-derived ceramide acts as a signaling molecule that can activate kinases involved in stress responses. Additionally its role in apoptosis links it to various stress signals where it facilitates the programmed cell death process. Interaction with proteins such as caspase and protein kinase C highlights its integration into signaling networks illustrating its regulatory significance.

Altered nSMase activity associates with cancers and neurodegenerative diseases.
