NSP3 also known as non-structural protein 3 is a protein involved in several mechanical processes within viral replication. This protein has a mass of roughly 219 kDa and is expressed in cells infected by certain viruses like the SARS-CoV-2 virus responsible for COVID-19. Mechanically NSP3 functions by interacting with host cell translation machinery and is important for the processing of viral polyproteins. NSP3 contains multiple domains including a papain-like protease (PLpro) domain that play roles in modifying host and viral proteins to assist in viral replication and pathogenesis.

This protein plays significant roles in immune evasion and replication of viral RNA. NSP3 is a component of the viral replication transcription complex which collaborates with other non-structural proteins such as NSP4 and NSP6.
