NUBPL also known as nucleotide-binding protein-like and IND1 is a mitochondrial matrix protein with a molecular mass of approximately 34 kDa. It is expressed in various tissues being notably present in high-energy demand tissues like heart and skeletal muscle. Mechanically NUBPL plays a critical role in the assembly of complex I of the mitochondrial respiratory chain by aiding the incorporation of Fe-S clusters which are essential for catalytic activity.
Biological function summary

NUBPL functions as a chaperone-like protein ensuring proper assembly of complex I which is part of the mitochondrial respiratory chain. NUBPL operates as a component of the mitochondrial iron-sulfur (Fe-S) cluster assembly complex. It directly interacts with Fe-S protein synthesis an essential part of cellular respiration. The precise regulation of this complex facilitates energy production within cells by supporting efficient electron transport.

Pathways

NUBPL is integral to the mitochondrial oxidative phosphorylation pathway a process that generates ATP the main energy currency of cells. It associates with the iron-sulfur (Fe-S) cluster biogenesis pathway working alongside proteins such as ISCU and NFS1. These interactions ensure the effective transfer and stabilization of Fe-S clusters necessary for mitochondrial function and metabolic processes.

NUBPL mutations have been linked to mitochondrial complex I deficiency which can lead to a spectrum of neurodegenerative conditions including Leigh syndrome. The disease often involves interactions with proteins such as NDUFS4 another complex I assembly factor. Additionally possible links may exist between NUBPL dysfunction and mitochondrial diseases characterized by impaired energy production and accumulation of metabolic intermediates highlighting the protein's importance in maintaining cellular energy homeostasis.
