Supplementary information

The Nucleolar protein 14 (NOP14) also known as Cirhin or BCDS is a protein with a mass of approximately 90 kDa. It functions mechanically as an important component of ribosome biogenesis specifically during the early stages of 90S pre-ribosomal particle assembly. NOP14 is expressed largely within the nucleolus a sub-nuclear structure that is actively engaged in ribosomal RNA synthesis and ribosome assembly. Additionally NOP14 interacts with other nucleolar proteins and participates actively in the maturation of small subunit ribosomal RNA.
Biological function summary

NOP14 plays a significant role in the assembly of ribosomal subunits forming part of the small subunit (SSU) processome complex. This complex is vital for the processing of the 35S pre-rRNA into the 18S rRNA an important step in the formation of the 40S small ribosomal subunit. The SSU processome involves various proteins and small nucleolar RNAs emphasizing NOP14’s significance in both the structure and function within this large nucleolar complex. The integrity of ribosome production relies heavily on the precise actions of NOP14 within this multifaceted process.

Pathways

NOP14 participates in ribosome biogenesis a critical cellular pathway affecting protein synthesis. It involves significant interactions with the U3 small nucleolar RNP and other ribosome-associated proteins. Another essential pathway that NOP14 impacts is the translation initiation pathway where ribosome assembly begins conjoined efforts with other proteins like NOP56 and NOP58 influencing eukaryotic cell growth and division. These interactions highlight NOP14's interconnected role within cellular proliferation and growth regulation.

NOP14 is linked to North American Indian childhood cirrhosis (NAIC) a hepatological disorder. Mutations within the CIRH1A gene which encodes NOP14 disrupt normal ribosomal assembly leading to hepatic dysfunction. Additionally NOP14 associates with disorders related to ribosomopathies where defects in ribosomal function lead to diverse developmental anomalies and pathological conditions. An understanding of NOP14’s interaction with proteins like RPS19 in these contexts can provide insights into therapeutic targets for these ribosome-associated disorders.
