This data was developed using ab134164, the same antibody clone in a different buffer formulation.

Lanes 1 - 3 : Merged signal (red and green). Green - ab134164 observed at 100 kDa. Red - loading control, ab8245, observed at 37 kDa.

ab134164 was shown to specifically react with Nucleolin in wild-type HeLa cells as signal was lost in Nucleolin knockout cells. Wild-type and Nucleolin knockout samples were subjected to SDS-PAGE. The membrane was blocked with 3% Milk. ab134164 and ab8245 (Mouse anti GAPDH loading control) were incubated overnight at 4°C at 1/10000 dilution and 1/20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

Nucleolin also known as C23 is a multifunctional protein mainly found in the nucleolus with a molecular weight of approximately 100 kDa. It plays a mechanical role in ribosomal RNA (rRNA) synthesis and ribosome biogenesis. Nucleolin is widely expressed in proliferating cells including HeLa culture and shows higher expression in cancer cells compared to normal cells. The nucleolin protein is involved in various nucleolar processes like the modulation of chromatin structure and nucleocytoplasmic transport.
Biological function summary

The nucleolin molecular weight protein contributes to regulating cellular processes such as chromatin remodeling rRNA transcription and mRNA stability. Nucleolin often forms complexes with various other proteins and nucleic acids enhancing its functionality within the cell nucleus. Nucleolin is the major nucleolar protein of growing eukaryotic cells. It is found associated with intranucleolar chromatin and pre-ribosomal particles. It induces chromatin decondensation by binding to histone H1. It is thought to play a role in pre-rRNA transcription and ribosome assembly. May play a role in the process of transcriptional elongation. Binds RNA oligonucleotides with 5'-UUAGGG-3' repeats more tightly than the telomeric single-stranded DNA 5'-TTAGGG-3' repeats.
