The SARS-CoV-2 Nucleoprotein often referred to as the N protein plays a central role in the life cycle of the virus. It has a molecular mass of approximately 45 to 50 kDa and is encoded by the N gene. The N protein is expressed in the cytoplasm of infected cells. It forms the viral ribonucleoprotein complex by binding to the viral RNA genome ensuring its proper packaging into new particles. The N protein supports viral replication and assembly through interactions with other viral proteins.
Biological function summary

The N protein interacts with several host cellular components to facilitate viral replication. It is not part of a large complex but forms homodimers which are essential for its function. Through these interactions the N protein disrupts normal cellular processes to favor viral replication. The N protein can modulate the host's immune response by interfering with signal transduction pathways this helps the virus evade detection.

Pathways

Many cell processes are hijacked by the N protein. It is involved in the viral replication and transcription complex which includes interacting with the host's translation machinery. The N protein associates with proteins like RNA polymerase to ensure efficient viral genome replication. It also affects the intracellular transport and assembly pathways of the virus critical for the production of infectious virions.

The N protein has a significant role in COVID-19. Its presence is often used as a marker for infection and is a target for diagnostic assays. The N protein can trigger an excessive immune response leading to cytokine storms associated with severe acute respiratory distress syndrome (ARDS) in COVID-19 patients. It is connected to other viral proteins such as the spike protein which is involved in the virus's ability to enter host cells compounding its impact on disease progression.
