Nucleoside-diphosphate kinase (NDPK) also known as nucleoside-diphosphate kinase NM23 and NDP kinase mechanically catalyzes the exchange of phosphate groups between different nucleoside diphosphates and triphosphates. This protein weighs approximately 17 kDa and is expressed in various tissues including liver heart and brain. Its catalytic function relies on a ping-pong mechanism where a high-energy phosphoryl transfer occurs important for maintaining the balance of cellular nucleotide pools.
Nucleoside-diphosphate kinase facilitates essential cellular processes by regulating the levels of nucleoside triphosphates key building blocks for nucleic acid synthesis. It is often found as part of a larger complex composed of hexameric structures in cells impacting cellular energy homeostasis and metabolism. The protein's role extends to involvement in cellular signaling differentiation and proliferation contributing to its importance in cellular regulation.
The role of nucleoside-diphosphate kinase extends to critical biological pathways such as the nucleotide metabolism and cell signaling pathways where it ensures the equilibrium of nucleoside triphosphates used in DNA and RNA synthesis. NDPK interacts with other proteins particularly linking with kinases and phosphatases to perform its regulatory functions. This protein integrates closely with the MAPK signaling pathway influencing cellular responses to external stimuli and cellular growth.
Issues with nucleoside-diphosphate kinase have been linked to conditions such as cancer and neurodegenerative diseases. Mutations or aberrant expression of NDPK can impact its function leading to alterations in cellular processes that drive oncogenesis. For example its altered interaction with proteins like p53 in cancer can impair tumor suppressor functions facilitating tumorigenesis. Additionally its dysregulation has associations with neurodegenerative diseases where it may affect neuronal survival and function.
