mRNA decapping enzyme that specifically removes the nicotinamide adenine dinucleotide (NAD) cap from a subset of mRNAs by hydrolyzing the diphosphate linkage to produce nicotinamide mononucleotide (NMN) and 5' monophosphate mRNA (PubMed:31101919, PubMed:31875550). The NAD-cap is present at the 5'-end of some RNAs; in contrast to the canonical N7 methylguanosine (m7G) cap, the NAD cap promotes mRNA decay (PubMed:31101919). Preferentially acts on NAD-capped transcripts in response to nutrient stress (PubMed:31101919). Also acts on free nicotinamide adenine dinucleotide molecules: hydrolyzes NAD(H) into NMN(H) and AMP, and NADPH into NMNH and 2',5'-ADP (PubMed:12790796). May act to regulate the concentration of peroxisomal nicotinamide nucleotide cofactors required for oxidative metabolism in this organelle (PubMed:12790796). Regulates the levels of circadian clock components PER1, PER2, PER3 and CRY2 in the liver (By similarity).

NUDT12 also known as Nudix hydrolase 12 plays a role in hydrolyzing nucleoside diphosphates. It mainly acts on ADP-ribose a product from NAD+ metabolism. The molecular mass of NUDT12 is approximately 43 kDa. Expression of NUDT12 occurs significantly in liver tissues and to some extent in other tissues as well. It contributes to maintaining nucleotide pools ensuring cellular homeostasis by freeing the cells from non-canonical nucleotides that can be harmful.
NUDT12 regulates cellular functions by processing NAD+ metabolites. It forms part of a complex involved with enzymes that manage nucleotide hydrolysis. By removing non-standard nucleotides from the internal environment it prevents harmful incorporation into nucleic acids. Its activity supports processes like DNA repair safeguarding genome integrity which is important for normal cell function.
NUDT12 functions within NAD+ metabolism and the nucleotide salvage pathway. It is part of a group of Nudix hydrolases that includes proteins like NUDT1. These proteins collectively aid in nucleotide management and energy homeostasis. By collaborating in these pathways NUDT12 ensures metabolic processes proceed without interference from aberrant molecules.
