Fatty acyl-coenzyme A (CoA) diphosphatase that hydrolyzes fatty acyl-CoA to yield acyl-4'-phosphopantetheine and adenosine 3',5'-bisphosphate (By similarity). Mediates the hydrolysis of a wide range of CoA esters, including choloyl-CoA and branched-chain fatty-acyl-CoA esters and at low substrate concentrations medium and long-chain fatty-acyl-CoA esters are the primary substrates (By similarity). Highest activity seen with medium-chain acyl-CoA esters and higher rates of activity seen with the unsaturated acyl-CoA esters compared with the saturated esters (By similarity). Exhibits decapping activity towards dpCoA-capped RNAs in vitro (By similarity).

NUDT19 also known as Diphosphatase NUDT19 is a protein that functions by hydrolyzing nucleoside diphosphates which are involved in various biochemical processes such as cell signaling and metabolism. It carries a molecular mass of approximately 22 kDa. NUDT19 mostly expresses in the kidneys and liver playing important roles in detoxification and energy metabolism in these tissues.
NUDT19 is involved in regulating intracellular levels of Coenzyme A and its derivatives through its enzymatic action. By breaking down these molecules it helps maintain cellular metabolic balance. While it does not function as part of a larger complex its activity directly impacts the availability of metabolic substrates required for energy production and other cellular functions.
NUDT19 plays a critical role in the Coenzyme A biosynthesis pathway. It contributes to the regulation of this pathway by controlling the levels of intermediates through its enzymatic activity. This protein links to other key enzymes such as ACSM3 which plays a part in the catabolism of fatty acids. These interactions highlight NUDT19’s involvement in lipid metabolism and energy balance.
NUDT19 has connections to metabolic disorders such as diabetes and fatty liver disease. Abnormal activity or expression of NUDT19 may lead to disruptions in energy metabolism contributing to the pathology of these conditions. Its role in managing Coenzyme A derivatives also ties it to SIRT5 a protein implicated in metabolic regulation and stress responses. Understanding NUDT19’s function could lead to insights into therapeutic strategies for related metabolic disorders.
