NUP88 mutants have distinct effects on NUP88 nucleoporin partners.(A) GFP-trap assays to study the effects of NUP88 mutants on its interaction with NUP214, NUP62 and NUP98. GFP and GFP-NUP88 fusion proteins were transiently expressed in HeLa cells. After 48 h, GFP proteins and associated factors were recovered from cell lysates and probed by Western blot analysis using antibodies against Nup214, NUP62, and NUP98. Successful transfection and expression of the proteins was confirmed by probing with antibodies against GFP. GFP proteins had the following size : GFP-NUP88 WT, D434Y, and E634del : ~115 (88 + 27) kDa; GFP-NUP88 R509* : 83 (56 + 27) kDa; GFP : 27 kDa. (B-D) HeLa cells were transiently transfected with GFP-constructs of the respective NUP88 mutant and fixed and stained after 48 h for immunofluorescence microscopy using (B) anti-NUP214, (C) anti-NUP62, and (D) anti-NUP98 antibodies. Only NUP88 E634del affects NUP62 association with NPCs. Scale bars : 10 μm.

The nucleoporin 214 (NUP214) also known as CAN is an essential component of the nuclear pore complex (NPC) which facilitates the transport of molecules between the nucleus and the cytoplasm. NUP214 has a molecular mass of approximately 214 kDa a characteristic that partially defines its naming. It expresses widely in various cell types reflecting its fundamental role in cellular transport processes. Within the NPC NUP214 partners with other nucleoporins to form critical subunits that ensure efficient nucleocytoplasmic transport.
Biological function summary

NUP214 participates in the assembly and function of the NPC by regulating cargo movement. This protein acts as a docking site for transport receptors aiding in the import and export of macromolecules. NUP214 exhibits a part in the nuclear pore complex interacting dynamically with other nucleoporins like NUP88 and NUP62. Its biological role maintains nuclear and cytoplasmic homeostasis which is important for normal cell function and division influencing gene expression and cellular responses to stimuli.

Pathways

NUP214 plays a significant role in nucleocytoplasmic transport mechanisms and is integrated into the Ran GTPase pathway. This pathway ensures that proteins and RNA efficiently move across the nuclear envelope fulfilling specific cellular functions. NUP214's functional relationships include interactions with importin and exportin proteins which recognize nuclear localization and export signals on cargo molecules. These interactions are vital for the cell cycle and signal transduction pathways highlighting NUP214's involvement in important cellular activities.

NUP214 often associates with certain forms of leukemia such as acute myeloid leukemia (AML). Aberrations in NUP214 like translocations or amplifications can lead to malignancies through disrupted nucleocytoplasmic transport and abnormal gene expression. The fusion protein generated from NUP214-ABL1 resulting from a chromosomal translocation directly involves in leukemogenesis by activating abnormal signaling pathways contributing to oncogenesis. Understanding NUP214's role in these disorders provides insight into potential therapeutic targets for treatment.
