Paraffin-embedded human testis tissue stained for NUP93 using ab238735 at 1/400 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

NUP93 also known as nucleoporin 93 is a protein with a molecular mass of approximately 93 kDa. It forms a part of the nuclear pore complex (NPC) specifically contributing to its structural integrity. NUP93 can be found in the nuclear envelope of eukaryotic cells where it aids in the proper functioning of the NPC. The protein ensures that macromolecules can efficiently transport between the nucleus and the cytoplasm therefore maintaining cellular function.
Biological function summary

NUP93 plays an important role in the assembly and stability of the nuclear pore complex by interacting with other nucleoporins. It is a component of the core scaffold structure interacting with proteins like NUP205 and NUP188 and helps in the spatial arrangement and functionality within the complex. Through these interactions NUP93 contributes to selective transport acting as gatekeeper to regulate traffic into and out of the nucleus.

Pathways

NUP93 influences various cellular processes by affecting nuclear transport pathways such as the nucleocytoplasmic transport pathway. Through its connectivity in the nuclear pore complex it affects the transit of molecules like messenger RNA (mRNA) and ribonucleoprotein complexes. NUP93 is associated with proteins such as importin-beta which is involved in the import of proteins with nuclear localization signals thereby ensuring precise nuclear-cytoplasmic communication.

Mutations or dysregulation of NUP93 have associations with nephrotic syndrome and some forms of cancer. Nephrotic syndrome can result from altered nuclear pore function where NUP93 links with proteins like podocin affecting kidney filtration efficiency.
