Activity summary

Nuclear receptor-related 1 protein (Nurr1) is a transcription factor weighing approximately 66 kDa. Scientists also know Nurr1 as NR4A2 and it belongs to the NR4A nuclear receptor family. Nurr1 is mainly expressed in the central nervous system where it plays a significant role in neuronal development and maintenance. Despite lacking a defined ligand-binding activity Nurr1 regulates gene expression directly by binding to DNA.

Biological function summary

Nurr1 functions as a critical regulator of dopaminergic neurons in the brain. It is vital for the survival and differentiation of these neurons which are important for motor control and behavioral responses. Nurr1 partners with other transcription factors often functioning in complexes that modulate brain-derived neurotrophic factor (BDNF) signaling. It influences the transcription of genes involved in inflammatory responses and neuroprotection reflecting its importance in maintaining neural health.

Pathways

Nurr1 integrates into key biological processes involving dopaminergic signaling and neuroinflammation. Nurr1 directly participates in the Wnt signaling pathway which plays a role in neuronal development and synaptic function. It also interacts with proteins such as Pitx3 and LMX1B within these pathways contributing to the regulation of dopaminergic neuron function and survival ultimately affecting broader central nervous system activity.

Associated diseases and disorders

Nurr1 has strong associations with neurological conditions such as Parkinson’s disease and schizophrenia. Its impaired function or altered expression can lead to dopaminergic neuron degeneration a hallmark of Parkinson's disease. Research indicates Nurr1's interaction with parkin a protein implicated in the disease's pathology further emphasizes its importance. In schizophrenia altered Nurr1 activity affects synaptic regulation and cognitive function potentially relating to disrupted dopaminergic and BDNF pathways.
