Enrichment of endogenous Oct-1 protein at specific gene loci using ab308043 : Chromatin Immunoprecipitation (ChIP) was performed using ab308043 at 4 μg on sheared chromatin from 2 million HeLa cells. Normal Rabbit IgG was used as a negative IP control. The purified DNA was analyzed by qPCR with PCR primer pairs over Promoters of HIST2H2BE, POLR2A (positive) and SAT2 satellite repeats (negative). Data is presented as fold enrichment of the antibody signal versus the negative control IgG using the comparative CT method.

Western blot analysis was performed on modified whole cell extracts (1% SDS) of HeLa (Lane 1), Hep G2 (Lane 2), A-431 (Lane 3), Ramos (Lane 4), HEK-293 (Lane 5). HCT 116 (Lane 6) and NTERA-2 (Lane 7). The blot was probed with ab308043 at 1 : 250 dilution and detected by chemiluminescence using a Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate at 1 : 4000 dilution. A ~80 kDa band corresponding to Oct-1 was observed across the cell lines tested.

Knockdown of Oct-1 was achieved by transfecting HeLa cells with Oct-1 specific siRNA. Western blot analysis was performed using modified whole cell extracts (1% SDS) from Oct-1 knockdown cells (Lane 3), non-specific scrambled siRNA transfected cells (Lane 2) and untransfected cells (Lane 1). The blot was probed with ab308043 at 1/1000 dilution and a Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate at 1/4000 dilution.

Oct-1 also known as POU domain class 2 transcription factor 1 (POU2F1) is a transcription factor characterized by its POU-specific domain. The molecular weight of Oct-1 is approximately 76 kDa. It is expressed widely across different tissues with high levels found in lymphoid organs and epithelial cells. It binds to octamer motifs in the promoters of target genes regulating their transcription. Oct-1 plays a role in the cell cycle development and immune response.
Biological function summary

Oct-1 participates in the regulation of gene expression by influencing RNA polymerase II.
