Secreted scaffold protein that plays an essential role in dorsoventral patterning during early development. Stabilizes axial formation by restricting chordin (CHRD) activity on the dorsal side. Acts by facilitating the association between the tolloid proteases and their substrate chordin (CHRD), leading to enhance chordin (CHRD) degradation (By similarity). May have matrix-related function involved in placental and embryonic development, or play a similar role in other physiological processes.

OLFML3 also known as Olfactomedin-like protein 3 is a secreted glycoprotein with a molecular mass of approximately 50 kDa. It belongs to the olfactomedin domain-containing protein family. This protein mainly expresses in various tissues including the eyes brain and heart. It often emerges in areas involved in development and growth contributing to its functional significance in different physiological contexts.
OLFML3 plays an integral role in cell adhesion and development processes. It acts as a regulatory molecule facilitating cell communication in tissue development. Studies suggest its involvement in promoting angiogenesis the formation of new blood vessels thereby assisting in tissue repair and regeneration. OLFML3 may associate with components of the extracellular matrix suggesting its potential presence in larger protein complexes that modulate cellular environments.
OLFML3 significantly interacts in pathways related to angiogenesis and tissue remodeling. It plays a role in the VEGF (Vascular Endothelial Growth Factor) signaling pathway which is critical for angiogenesis. OLFML3 functions alongside proteins like VEGFR2 enhancing the process of new blood vessel formation in response to hypoxic conditions or injury which is essential in maintaining tissue homeostasis and repair.
Researchers link OLFML3 to cardiovascular diseases and cancer progression. An overexpression of OLFML3 has been observed in certain cancers where it may promote tumor angiogenesis aiding in cancer growth and metastasis. Additionally disruptions in its function could associate with abnormal heart developments implicating its relationship with proteins like MMPs (matrix metalloproteinases) that participate in matrix remodeling processes involved in both tumor and cardiovascular pathology.
