ORC1 also known as Origin Recognition Complex subunit 1 is an important component involved in the initiation of DNA replication. It forms part of a larger protein complex that plays a role in recognizing the starting points or origins for replication to begin. ORC1 has a molecular mass of approximately 97 kDa and is expressed in various cell types throughout the body but is especially prominent in cells that actively divide. Its expression ensures that the replication machinery assembles correctly during the cell cycle.
Biological function summary

In terms of chromosomal DNA replication ORC1 plays a central role in forming the pre-replicative complex. This protein is a component of the ORC complex which includes several other subunits responsible for facilitating replication initiation across eukaryotic cells. ORC1 through interactions with additional proteins like CDC6 and CDT1 helps load the MCM helicase onto DNA which is essential for DNA unwinding before replication.

Pathways

The replication origins and ORC1's role in them tie it to the DNA replication and cell cycle pathways. In the DNA replication pathway ORC1 serves as the initial marker for DNA segment to act as a template for the replication machinery. Additionally its relationship with CDC6 links it to the cell cycle regulatory pathways orchestrating timely DNA replication to ensure that cell division occurs accurately and efficiently.

Defects or dysregulation in ORC1 are linked to Meier-Gorlin syndrome a condition characterized by growth delays and developmental abnormalities. Further irregularities in ORC1 function can also contribute to certain cancers due to uncontrolled cell division. Within these disorders connections between ORC1 and proteins such as CDT1 emphasize its role in ensuring orderly cell cycle progression whose disruption can lead to disease pathogenesis.
