Supplementary information

ORC6L also known as Origin Recognition Complex Subunit 6 is a protein that plays a mechanical role in the initiation of DNA replication. It weighs approximately 30 kDa and localizes in the nucleoplasm of cells. ORC6L is expressed in various tissues with higher expression observed in testis pancreas and thymus. It is an important component ensuring the proper functioning and regulation of DNA replication origins.
Biological function summary

DNA replication relies on ORC6L as part of the origin recognition complex (ORC) a multi-protein assembly essential for marking the origins of replication. The ORC complex of which ORC6L is a vital member initiates the assembly of pre-replicative complexes important for cell cycle progression. By binding to specific DNA sites ORC6L helps recruit other initiation factors ensuring that DNA replication occurs only once per cell cycle maintaining genomic stability.

Pathways

Different phases of the cell cycle require ORC6L's role in the DNA replication pathway. This protein's function connects with other components of the ORC such as ORC1 ORC2 and ORC3 to control replication initiation. Furthermore ORC6L interacts with proteins such as CDC6 and MCM integrating into the DNA replication licensing pathway which ensures the cell's readiness for DNA synthesis and subsequent divisions.

Mutations or dysregulation of ORC6L have implications for genomic instability and diseases like Meier-Gorlin syndrome. This condition characterized by growth retardation and skeletal anomalies often links to other ORC components including ORC1 and ORC4 illustrating its involvement in developmental abnormalities. Additionally cancer research indicates the possible connection between ORC6L dysfunction and tumorigenesis due to unchecked cell proliferation emphasizing its importance in maintaining a balanced cell division process.

Target data

Component of the origin recognition complex (ORC) that binds origins of replication. DNA-binding is ATP-dependent. The specific DNA sequences that define origins of replication have not been identified yet. ORC is required to assemble the pre-replication complex necessary to initiate DNA replication. Does not bind histone H3 and H4 trimethylation marks H3K9me3, H3K27me3 and H4K20me3.
