In Western blot, ab273435 was shown to bind specifically to ORF7a Protein. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in fluorescent western blot (TBS-based) blocking solution before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Mouse IgG H&L (IRDye® 800CW) preabsorbed (ab216772) and Goat anti-Rabbit IgG H&L (IRDye® 680RD) preabsorbed (ab216777) at 1/20000 dilution.

The ORF7a protein also known as accessory protein 7a acts as an important virulence factor in SARS-CoV-2 the virus causing COVID-19. The protein has a molecular mass of approximately 12 kDa and is expressed in the endoplasmic reticulum. It extends into the lumen where it can bind to proteins and other molecules. ORF7a is known to interact with different host proteins manipulating cellular machinery to aid viral replication and pathogenesis.
Biological function summary

Functions of the ORF7a protein extend to immune system inhibition. It can interfere with type I interferon signaling reducing the host's antiviral response. As part of viral assembly ORF7a contributes to the efficient packaging and release of viral particles. It does not form a complex with other viral proteins but acts independently to execute its functions within the infected host cell.

Pathways

Roles of the ORF7a protein integrate into immune escape and virion production processes. It participates in immune evasion pathways by inhibiting signaling pathways that interfere with interferon production like the JAK-STAT pathway. Additionally it has associations with other SARS-CoV-2 proteins like ORF3b which jointly compromise host cell functions and enhance virus survival.

The ORF7a protein has strong connections to the manifestation of COVID-19-related symptoms. The inhibition of interferon signaling contributes to respiratory distress observed in severe cases of COVID-19. Studies also indicate links to the inflammatory responses involving other viral proteins such as the N protein which together can exacerbate the severity of SARS-CoV-2 infections.
