What is a lysosome?
Lysosomes are membrane-bound organelles essential for cellular waste processing and recycling. LAMP1 and LAMP2 (Lysosome-Associated Membrane Proteins 1 and 2) are widely used markers for the lysosomal membrane, often targeted in immunofluorescence and cell imaging to visualize lysosomal distribution and abundance. A key player in lysosomal enzyme trafficking is the mannose-6-phosphate receptor (M6PR), which resides primarily in the trans-Golgi network and endosomes. M6PR recognizes lysosomal enzymes tagged with mannose-6-phosphate and directs them to lysosomes, ensuring proper delivery of hydrolases.

What is an autophagosome?
Autophagosomes are double-membraned vesicles that play a central role in autophagy, a cellular degradation pathway that recycles cytoplasmic components. One of the most widely used markers is LC3 (microtubule-associated protein 1A/1B-light chain 3), particularly its lipidated form, LC3-II, which integrates into the autophagosomal membrane and is essential for autophagosome elongation. Another key marker is p62/SQSTM1, a cargo adaptor protein that links ubiquitinated substrates to LC3 and is degraded upon autophagy completion, making it a useful indicator of autophagic flux.
