OVCA1 also known as DPH1 or DPH2L1 is a protein encoded by a gene located in human chromosome 11p11.2. The protein weighs approximately 42 kDa. OVCA1 is involved in the early steps of diphthamide biosynthesis where it functions by interacting with its protein partners. It shows expression in various tissues with higher levels in the ovary and heart. OVCA1's role gets linked to cellular processes that impact cell growth and survival.
Biological function summary

This protein plays important roles in cellular proliferation and maintenance. OVCA1 is a part of the DPH1-DPH2 complex that facilitates the modification of eukaryotic elongation factor 2 (eEF2) through diphthamide synthesis. The process of 2-[3-carboxamido-3-(trimethylammonio)propyl]histidine diphthamide biosynthesis is essential for proper cellular protein translation. OVCA1 helps regulate the diphthamide biosynthesis pathway's first step thereby impacting the eEF2’s functionality which is important for protein synthesis and cell division.

Catalyzes the first step of diphthamide biosynthesis, a post-translational modification of histidine which occurs in elongation factor 2 (PubMed : 30877278). DPH1 and DPH2 transfer a 3-amino-3-carboxypropyl (ACP) group from S-adenosyl-L-methionine (SAM) to a histidine residue, the reaction is assisted by a reduction system comprising DPH3 and a NADH-dependent reductase (By similarity).
